sgRNA_gpm_STREPTO
This part encodes a single guide RNA targeting the gpm gene encoding a 2,3-bisphosphoglycerate-dependent phosphoglycerate mutase (short name: phosphoglyceromutase) in Streptomyces. It has been designed to target different strains such as S. ambofaciens ATCC 23877, S. bottropensis ATCC 25435, and S. venezuelae ATCC 10712.
Usage and Biology
A decrease of gpm expression has been shown as required to allow the implementation of the Calcin cycle in E. coli (Antonovsky et al., Cell, 2016). This part could therefore be useful to implement a CO2 fixation module in Streptomyces.
We have engineered genetic tools via the Crispr-dCas9 system to switch off gpm and force the appearance of the Calvin cycle in their primary metabolism.
